Do you have any positive control for your ROS production from Raw 264.7 ? Such as LPS or any other chemicals.
Generally, if you start with any assay you are not good at, try with the positive control is necessary.
Also, sometimes check your cell under microscope, make sure that the cells have some response to reduce the possibility that your spectrometer does not work well.
